چکیده

Statistics of mitochondrial association with spindle poles in wild type and mto1∆ (Figures 1D and 2B) Out of the 35 imaged wild-type cells, 18 cells showed constant mitochondrial association (red bar in Figure 1D), 6 showed at least one spindle pole free from mitochondria for at least 5 minutes (green bar in Figure 1D), 6 displayed temporary mitochondrial detachment (not included in the comparison), and for 5 cells the association periods could not be determined accurately (not analyzed). The 6 free spindles were identified as free based on the observation that the distance between a spindle pole and the nearest mitochondrion was larger than ~200 nm. Out of the 22 imaged mto1∆ cells, 10 cells showed constant association with either a mitochondrial network or a small globular mitochondrion (red bar in Figure 2B), 9 showed at least one spindle pole free for at least 5 minutes (green bar in Figure 2B), and 3 displayed temporary association (not included in the comparison). Out of the 9 free spindles, 8 were identified as free based on the observation that the distance between a spindle pole and the nearest mitochondrion was larger than ~200 nm, and the remaining one based on the observation that the movements of the spindle and the adjacent mitochondria were uncorrelated.

Mitochondrial positioning in fission yeast is driven by association with dynamic microtubules and mitotic spindle poles.

Microtubules mediate mitochondrial distribution in the yeast Schizosaccharomyces pombe and many higher eukaryotic cells. In higher eukaryotes, kinesin motor proteins have been shown to transport mitochondria along microtubules, but the nature of the mitochondria-microtubule interactions in S. pombe has not been explored. By time lapse, total internal reflection fluorescence microscopy, or spinn...

Polo-like kinase 1 facilitates chromosome alignment during prometaphase through BubR1.

Plk1, an evolutionarily conserved M phase kinase, associates with not only spindle poles but also kinetochores during prometaphase. However, the role of Plk1 at kinetochores has been poorly understood. Here we show that BubR1 mediates the action of Plk1 at kinetochores for proper chromosome alignment. Our results show that BubR1 colocalizes with Plk1 at kinetochores of unaligned chromosomes and...

Clathrin recruits phosphorylated TACC3 to spindle poles for bipolar spindle assembly and chromosome alignment.

Transforming acidic coiled-coil-containing protein 3 (TACC3) has been implicated in mitotic spindle assembly, although the mechanisms involved are largely unknown. Here we identify that clathrin heavy chain (CHC) binds specifically to phosphorylated TACC3 and recruits it to spindle poles for proper spindle assembly and chromosome alignment. Phosphorylation of Xenopus TACC3 at serine 620 (S620) ...

Asymmetric Loading of Kar9 onto Spindle Poles and Microtubules Ensures Proper Spindle Alignment

Spindle alignment is the process in which the two spindle poles are directed toward preselected and opposite cell ends. In budding yeast, the APC-related molecule Kar9 is required for proper alignment of the spindle with the mother-bud axis. We find that Kar9 localizes to the prospective daughter cell spindle pole. Kar9 is transferred from the pole to cytoplasmic microtubules, which are then gu...

O-9: Spindle and Chromosome Configurations of Human Oocytes Matured In vitro in Two Different Culture Media

Background: In vitro maturation can have deleterious effects on spindle formation and proper chromosome alignment in human oocytes and can be profoundly affected by culture conditions.
